Very important and differentiating reactions! A positive reaction is a colour change to blue. Particularly cells of the inner mantle portions can show such a reaction. A patchy distribution of this reaction can frequently be observed. Be sure that you do not misinterpret a similar colour change of root cell remnants on your mantle preparation as a positive reaction. When applying guaiac solution, the water around the mantle piece must be almost completely soaked off, otherwise the reagent will not get in contact with the hyphae and, in additon, an emulsion will make observations difficult.
